They have not used leather for the pumps in probably 30 years. A 424 will have a nitrile seal. If it is not pumping well it probably needs a couple of drops of oil.
It is not smaller. The dimensions shown earlier show that the 110 is the same width and a bit longer than a Disco 5. It will be basically the same price point.
They stated in one of the previous announcements it would have an inline six in both petrol and diesel. The diesel would have a turbo plus an electric supercharger.
The new diesel on the F150 is the same engine used on current Discoveries and Range Rovers ( 3 litre V6 Powerstroke). The new Defender is using a different engine. A straight 6.
Land Rovers used the same hub nut size as Jeeps. Everybody used that size until the 70s. It is all long before the Range Rover.
The issue with TC is it takes power always to give traction. This only works in certain situations.
$45k ? That is crazy money. You should not need to pay half of that for a good truck. The high priced ones are mall queens. Parts are very cheap and they are simple to work on.
Robert Davis already makes all the parts needed for this engine conversion. You need a custom oil pan and pickup.

Back to the options. You won’t be happy with the TDI. The rest are complex conversions and costly to do properly. If you need to ask on the internet they are probably beyond your abilities.
