Contact your local driveline or gear shop. They should be able to calculate what the RPM will be at a certain MPH with different gear/tire/trans.
I regeared to 4.56 and my guy sent me the RPM at 65 MPH with 4.10, 4.56 and 4.88

Would probably be easier to buy an existing open or closed trailer that will fit what you're looking for and modify what's necessary from there i.e. axle, wheels, tires, etc.
